SANGER SEQUENCING

Sample preparation guide

DNA and primer quantities, packaging, and delivery for Standard and Pre-mixed Sanger orders.

These quantities apply to Sanger sequencing. For Whole Plasmid or other services, view Nanopore services or contact our team for preparation requirements.

BEFORE YOU PREPARE

Confirm which quantities to use.

Both the listed quantities and 2× quantities are shown below. Confirm with the laboratory which set to prepare for your submission.

Ask our team
01 / STANDARD

DNA and primer supplied separately

Submit DNA separately from any customer-supplied primer. In your order, select a universal primer or identify the primer you are providing. Label all containers to match the order.

Swipe the table sideways to see all quantities.

2× primer volume
10 µL

DNA amount is a mass in ng; primer volume is in µL. Use the quantities confirmed by the laboratory.

02 / PRE-MIXED

DNA and primer supplied together

Identify the primer used for each mixture in your order. The tables show DNA amount, primer amount, and total mixture volume.

Use the set confirmed by the laboratory: listed quantities or 2× quantities.

Swipe the table sideways to see all quantities.

Primer amount in pmol is different from primer concentration in pmol/µL. The volumes above are the total volumes of the DNA–primer mixtures.

03 / PRIMERS

Choose and identify your primers

Universal primers

Universal primers are available from our laboratory. Contact our team for the current list and availability.

Customer-supplied primers

Label each primer clearly and identify its corresponding samples in the order. For Pre-mixed submissions, identify the primer included in each mixture.

04 / PACKAGING

Label and package your samples

SINGLE TUBES

1.5 mL tubes with pop caps

Use 1.5 mL microcentrifuge tubes with pop caps, labeled to match the order form.

For orders of 24 samples or more, use 96-well plates instead of tube strips.

96-WELL PLATES

Normalize and seal

Normalize DNA and primer concentrations according to the applicable preparation requirements. Arrange samples vertically: A1–H1, then A2–H2, and continue by column.

Seal or cap the plate with strip lids to prevent leakage.

For plate submissions, use one primer arrangement:

  1. A single primer for all samples.
  2. A matching primer plate, aligned well-to-well with the DNA plate, when using more than one primer.
  3. Primer already mixed with the DNA in each well.
05 / ORDERING & DELIVERY

Submit your order and arrange delivery

  1. Submit your online order

Choose Standard or Pre-mixed, enter your sample and primer details, and submit the order.

  2. Match the labels to the order

Check that each tube or plate position matches your sample list. Include your order form or a printed order confirmation with the samples.

  3. Confirm the drop-off or pickup arrangement

Select a Dropbox or another available delivery method during checkout. Placement details and pickup schedules depend on the location; confirm the arrangement before sending your samples.
